To demonstrate that the promoter of interest only functioned in kidney cells, a transgenic line was established with beta-galactosidase under the control of the promoter. RT-PCR could only detect expression in the kidney of beta-galactosidase but detected G3PDH in all tissues examined. The promoter of interest could gel shift with rhAP-2, and rhAP-2 could protect a 45bp region of the promoter in a footprinting experiment. (0423)
